چکیده

Hepatocellular carcinoma (HCC) is the fifth common malignant tumor worldwide and has the characteristics of insidious onset, high malignancy, invasive fast-growth, high recurrence and poor prognosis. Currently, besides the alpha-fetoprotein (AFP), there are still no other markers for the accurate HCC diagnosis. Emerging evidence shows that pyruvate kinase isozyme splice variant 2 (PKM2) is highly expressed in various tumors, and is a master regulator of cancer cell metabolism. In the occurrence and development of HCC, PKM2 is involved in a variety of signal pathways, such as Hedgehog, STAT3, PI3K/AKT and EGFR. Whether PKM2 may serve as a target for HCC diagnosis and prognosis is still unclear. Herein, we reviewed the emerging evidence about the effect of PKM2 on the HCC, aiming to push forward clinical application of PKM2 in the early diagnosis and prognosis evaluation of HCC.

Status of serum alpha feto-protein (AFP) and midkine (MDK) levels in patients with hepatocellular carcinoma

Introduction: Hepatocellular carcinoma (HCC) is sixth common cancers worldwide and predominant in Asia and Africa. A number of evidence suggests a possible role of midkine (MDK) and α-fetoprotein (AFP) in the pathogenesis of hepatocellular carcinoma (HCC). Materials and methods: We studied MDK and AFP in patients with HCC or in healthy controls. MDK and AFP was measured by enzyme linked immuno...

PKM2 regulates Gli1 expression in hepatocellular carcinoma

Hedgehog (Hh) signaling and the pyruvate kinase isoenzyme M2 (PKM2 or M2-PK) are often involved in tumorigenesis and growth. Aberrant activation of Hh signaling is found in a variety of malignancies. In tumor cells, PKM2 determines whether glucose is used for the synthesis of cellular building blocks or the production of lactate for energy regeneration; it associated with the Warburg effect. Gl...

Co-expression of PKM2 and TRIM35 predicts survival and recurrence in hepatocellular carcinoma

The identification of prognostic markers for hepatocellular carcinoma (HCC) is needed for clinical practice. Tripartite motif-containing 35 (TRIM35) is a tumor suppressor of HCC. TRIM35 inhibits phosphorylation of pyruvate kinase isoform M2 (PKM2), which is involved in aerobic glycolysis of cancer cells. We found that expression of PKM2 was significantly increased in HCC tissues. This overexpre...

PKM2 regulates hepatocellular carcinoma cell epithelial-mesenchymal transition and migration upon EGFR activation.

Pyruvate kinase isozyme type M2(PKM2) was first found in hepatocellular carcinoma(HCC), and its expression has been thought to correlate with prognosis. A large number of studies have demonstrated that epithelial-mesenchymal transition (EMT) is a crucial event in hepatocellular carcinoma (HCC) and associated metastasis, resulting in enhanced malignancy of HCC. However, the roles of PKM2 in HCC ...

SH2 domain-containing phosphatase 1 regulates pyruvate kinase M2 in hepatocellular carcinoma

Pyruvate kinase M2 (PKM2) is known to promote tumourigenesis through dimer formation of p-PKM2Y105. Here, we investigated whether SH2-containing protein tyrosine phosphatase 1 (SHP-1) decreases p-PKM2Y105 expression and, thus, determines the sensitivity of sorafenib through inhibiting the nuclear-related function of PKM2. Immunoprecipitation and immunoblot confirmed the effect of SHP-1 on PKM2Y...
